So you turned the heated bed off in manual control and you changed the g code to M190 S0 and nothing still happens?

Does the printer home itself on the x,y and z axis?
Does repetier host show the extruder heating up OK?
If you manually extrude the filament in manual control does it come out of the print head?

Did you use a raft? I would recommend using a raft for each print.

I have a pair of tweezers I use.Once I do the manual extrusion before the print starts I use the tweezers to remove the filament strand. Otherwsie this can prevent afhesion as the print head drags a piece of PLA across the bed.

The benefit of a raft is if the filament doesnt stick right away it wont affect your print because it will just be the first part of the raft that doesnt stick.

I would also make sure the temp of the extruder is at 210-220 for PLA.
